The film revolves around the life of Prabhu (played by Mahesh Babu), a young NRI who returns to his hometown in Kerala, India. Prabhu is a devotee of Lord Krishna and regularly visits the local temple to seek blessings. There, he meets Oshana (played by Niharika Rai), a beautiful and vibrant young woman who is also a devotee of Lord Krishna.

The title Om Shanti Oshana plays on “Om Shanti” (peace/chants) and “Oshana” (a Malayalam colloquialism for a lighthearted, mischievous exclamation).
